You can choose either one. On the Dojo page Sakurai said that you can choose and the one you don't choose is captured. (Pretty much alluded to that).

This definitely has some replayablity! Think about it. You can choose to start with either Mario or Kirby. That means that the cut scenes will have different characters and may turn out a bit differently. Then you can choose which princess to save.

I would save Zelda also.

there is about 5 adventure mode ones though, on that site just let the videos run 2 the end to see more. There are the 2 you posted. a Mario & pit one. Kirby and peach one. And an amazing DK and diddy kong one havnt checked but sure they will all play after the links posted. =]

Yeah, definitely! I haven't looked much into the adventure mode yet, but I guess it'll be one of those games where the characters you choose, the people you beat and things you do effect the story. Which is awesome!
