You don't need to come across as something your not, be yourself.
If your shy, maybe asking her if she was joking would be a bit embarrasing, so see how she acts when your around her.
Maybe text her, Email etc, get to know her better?
Then once you feel comfortable enough with her, and you and pretty certain she's into you, ask her out.

Before you even consider the girl you have to look at your confidence issue. Why are you so unconfident? It sounds like you have every reason to be. If you got into a relationship and you didn't have confidence it'll cause all sorts of issues.

Once you've looked at that, then explain to the girl you're an idiot for passing on her and that you like her too but you were worried about getting into a relationship then offer to take her out, get to know her a little bit and see where you're at.

I am not sure how old you are but in any case I'd advise you not to rush into things.
