The blaze happened at a building used for asylum and immigration tribunals.
The fire is believed to be on the fifth floor of the Breams Building in Chancery Lane - used by The Special Immigration Appeals Commission (SIAC).
London Fire Brigade said eight fire engines were sent and there were no immediate reports of any people being trapped.
One eyewitness described a "huge black plume of smoke" rising into the sky as fire engines rushed to the scene.
Police cordoned off Chancery Lane at the junction with Fleet Street and stopped traffic from entering.
City of London police are at the scene.
