Hoshino (creator) said she based the Earl on Dorian Grey from the League of Extraordinary Men. I think she pick Dorian's power to be Earl's power. That is to live long over the century. Dorian Grey would never die, until his old picture been destroyed. I think the Earl's weakness is the same as Dorian's.
She also was a fan of dolls so the D stands for Dolls
Gray is a mixture of white so it means black = evil and white = good, this means nobody is too evil or too good representing the innocence, take sumans case into this.
So humans are the dolls of good and evil
Also D.Gray-Man stands for Dorian Gray the original weilder of the Crown Clown original excorcist the earls stage partner the one who killed him the first time
Also D.gray-man manga is on hold starting from next chapter due to someone reporting plagiarism.
