    With a designer program (Such as photoshop)
    and code it, or cheat using fireworks coder or something..

    Be a little more specific, Web2.0 basically means modern, so do a sort of Gradient based layout.

    Take a look at devode.net or last.fm they look a little web2.0, but if your going for a valenties Habbo, try using superlove images etc etc..
